Dependence of RAW cells on extracellular arginine to support NO productiona

RAW 264.7 cells were plated in RPMI devoid of L-arginine or in RPMI supplemented with L-arginine as indicated, then stimulated by the addition of LPS (100 ng/ml) and IFNγ (25 U/ml) at time zero. Total nitrite accumulated in the medium conditioned by 1 × 106 cells over the subsequent 18 hr is shown. Assays were performed in triplicate, and the experiment repeated at least three times. Data shown are mean ± SEM (n = 3 experiments).
