Glutamate release from striatal synaptosomes

Synaptosomes were labeled with [3H]-glutamate as described in Materials and Methods. Synaptosomes were preincubated with 100 mM L-nitroarginine methyl ester (L-NAME) or 10 nM FK506 for 5 min prior to release of [3H]-glutamate elicited by addition of 40 mM KCl. KCl depolarization release of glutamate is indicated by K+. Glutamate release was quantified as described in Materials and Methods. For NMDA-dependent release of glutamate, synaptosomes were washed in Mg-free NTR buffer and preincubated with L-NAME or FK506 for 5 min prior to addition of 300 mM NMDA and 100 mM glycine. After 2 min, release of glutamate was quantified as described in Materials and Methods. *Statistically significant differences between drug treatment and either KCl or NMDA treated samples (p < 0.05 using paired and unpaired Student’s t tests).
