Cardiac-specific transgene expression

Cardiac-specific transgene RNA expression in mouse lines 131 and 140 was determined by ribonuclease (RNase) protection assay. The probe was derived from the 5′ end of the transgene and results in approximately 300 and 200 nt protected fragments for the transgene and the endogenous mouse Myh mRNAs, respectively. Three micrograms of total RNA was assayed and then analyzed on a 5% denaturing Polyacrylamide gel. P, probe; Tg, transgene; Myh, mouse Myh; α, rat αMyh; nH, nontransgenic heart; H, heart; T, tongue; Sk, skeletal muscle; L, liver; S, spleen; K, kidney; Lg, lung; B, brain; U, uterus.
