Characterization of Th1 and Th2 cytokine profiles in human γδ and αβ T cell clones

RNA was isolated from γδ and αβ T cell clones designated as VM22-5 (αβ+, CD4+, CD8), VM3-3 (Vγ9/Vδl+, CD4, CD8), VM4-4 (Vγ9+/Vδ2+, CD4+, CD8), and VM9-3 (Vγ9+/Vδ2+, CD4, CD8+) 1 week after feeding with PHA, IL-2, and irradiated feeder cells (spontaneous) as described in Materials and Methods, or 1 week after HIV or mock infection. The RNA samples were then subjected to TH1 and TH2 RT-PCR. The size of 494 bp, 458 bp, 462 bp, 414 bp, 628 bp, 352 bp, and 285 bp PCR product bands represent IFN-γ, IL-2-, IL-4-, IL-5-, IL-6-, IL-10-, and IL-13-specific messages, respectively.
