RT-PCR analysis of V δ 1-J δ TCR expression in surgical samples of normal, noninvolved, and inflamed, involved intestine of individual patients with CD, UC, diverticulitis (DIV), or normal (NL) mucosa from various locations along the length of the intestine

Mucosal cDNA samples were amplified first with primers specific for Vδ1 and Cδ and the amplified products were gel purified then reamplified in a second PCR reaction with the same Vδ1 primer used for the first amplification and with a reverse primer specific for each of the four Jδ genes. Vδ1-Jδ PCR products were electrophoresed through 2% agarose gels, blotted, and probed with an internal 32P-labeled Vδ1-specific oligonucleotide probe. Autoradiography was carried out for 24 hr. Asterisks indicate Vδ-Jδ profile of noninvolved colonic mucosa.
