  1. Transformants were replica plated to Xgal galactose plates, and white colonies were picked and retested. Western analysis was performed to verify expression of full-length fusion proteins, and the phenotype was shown to be plasmid-linked by plasmid isolation and retransformation into the original host strain. Sequencing of the MDM2 coding region revealed four separate mutations: G58D, C77Y (isolated twice), and Δ106-115/+18.
