Flow cytometry analysis of chimeric mice

Approximately 2 months after reconstitution of irradiated F1 mice with parental bone marrow, the extent of chimerism was confirmed by staining peripheral blood lymphocytes from each mouse with FITC-labeled anti-H-2b and PE-labeled anti-H-2d antibodies (Pharmingen) or with isotype-matched control antibodies. Mice containing less than 4% residual host lymphocytes were used for studies. Prior to in vitro culture, spleen cells were stained as above to confirm chimerism. Representative dot plots corresponding to the three types of chimeric mice are shown.
