Immunoblotting reactivity of RA serum and MAbs AHF1 and AHF7 on low-salt extracts of cultured keratinocytes and normal human epidermis separated by two-dimensional isoelectrofocusing/SDS-PAGE

In the extracts from cultured keratinocytes (upper half), the RA serum and the AHF monoclonal antibodies stain a series of antigens exhibiting an S-shaped migration pattern (pI ranging from 5.5 to 7.3 and molecular weights ranging from 40 up to >200 kDa). In the extracts from normal epidermis (lower half), the RA serum and the AHF MAbs stain the comma-shaped neutral/acidic isoform of filaggrin (pI ranging from 6 to 7.4 and molecular weights ranging from 37 to 40 kDa).
