Fig. 1

In situ hybridization of Ang-2 mRNA in first trimester villi at 9 weeks gestation (A–F, H) and term placenta (G). (A) Dark field examination shows the intense silver grains over syncytiotrophoblast with some over cytotrophoblast. The distribution pattern is patchy with some trophoblast highly labeled, whereas others are negative, even in the same villus. Boxed region L indicates such a region. (B) Bright field examination of area shown in A. (C) Higher magnification view of the boxed section in A. (D) A nearby slide stained with anti-cytokeratin antibody to confirm the integrity of trophoblast layers. (E) Bright field view, at high magnification of the boxed section in B. (F) Cross section of a villus confirms that silver grains highly label syncytiotrophobast. (G) Darkfield examination of in situ hybridization with Ang-2 antisense probe in term placenta. Only diffuse silver grains were observed. (H) Serial section hybridized with Ang-2 sense probe is negative control. Scale bar, 50 µm for A, B, H and 25 µm for C, D, E, F, G.
