Dual regulation by laminin 1 to inhibit (via α6 integrins) and stimulate (via α-DG) β-cell differentiation. Blocking laminin-1 binding to α6 integrins by GoH3 antibody, or inhibiting PI3K, MEK1 or actin polymerization by wortmannin or Ly294002, PD98059 or cytochalasin D, respectively, significantly increases the number of total and β cells. In contrast, blocking laminin-1 binding to α-DG by IIH6 antibody or heparin decreases significantly the number of total and β cells. In the presence of heparin, the effect of GoH3 to enhance β-cell differentiation is abolished.
