Effects of ET-1 on cAMP accumulation in CHO cells bearing WT or mutant hETB receptors. Cells with WT receptor (j) or mutant hETB receptors G57S (□) and R319W (j) were treated or not with 1028 M ET-1. Increasing concentrations of ET-1, from 10212 to 102 6 M, had no effect on cAMP accumulation in cells transfected with G57S or R319W mutant hETB receptors (data not shown). The intracellular cAMP was determined by RIA, and the values are expressed as percentage of the basal cAMP accumulation. Results are the means 6 SEM of three independent experiments.
