Hypoxia does not activate NF-κB—using a consensus oligonucleotide probe. Macrophages incubated as previously for 30 min. and nuclear levels of NF-κB determined by EMSA using a consensus oligonucleotide probe (see methods) (9A). Hypoxia does not activate NF-κB activity above basal normoxic levels. LPS (1 µg/ml for 30 mins) strongly induces NF-κB activation. (n = 4, P < 0.01). A representative EMSA is shown, demonstrating that the concensus probe reveals two distinct NF-κB bands, of which the upper (arrowed) is inducible by LPS (9B).
