Functional exogenous p53 expression in JSQ-3 cells transfected by TfRscFv-LipA2-p53 immunolipoplex in vitro. JSQ-3 cells were co-transfected in triplicate by Tf-LipA-complexed plasmid pBP100, which contains the luciferase reporter gene under the control of a p53 responsive promoter (21), together with p53 plasmid complexed with TfRscFv-LipA2. TfRscFv-LipA2-p53 co-transfection gave 54-fold more p53-responsive luciferase activity than TfrscFv-LipA2-pVec (p = 0.0001) and 11-fold more than TfrscFv-LipA2 only (p = 0.0002), demonstrating that exogenous wtp53 protein was expressed functionally by this gene delivery system.
