NO represses IL-1β-induced ICAM-1 promoter activity. Human A549 cells were transiently transfected with an ICAM-1 promoter-luciferase construct containing the 1014 nucleotide sequence of the 5′ region flanking the human ICAM-1 gene together with a β-galactosidase reference plasmid. Transfected cells were incubated with 2000 U/ml IL-1β, 2000 U/ml TNF-α, and 200 U/ml IFN-γ for 8 h in the absence or presence of increasing concentrations of DETA/NO. β-galactosidase-normalized luciferase reporter gene activities show that increasing concentrations of DETA/NO decrease the cytokine-induced ICAM-1 promoter activity.
