Dot blot analysis of the purified membrane preparation from platelets using biotin-ylated HK, factor XII, thrombin, and antibody to GPIb. Proteins purified from the HK affinity column (lane 1) and anti-glycocalicin antibody affinity column (lane 2) or purified IgG (lane 3 in E and F) were spotted onto the nitrocellulose membrane and probed with biotinylated HK (A,B) and biotinylated Factor XII (C,D). A and C are with zinc while B and D are without zinc. Binding of biotinylated thrombin (E) and anti-GPIb antibody (F) are shown in the absence of zinc.
