From: NO-Evoked Macrophage Apoptosis Is Attenuated by cAMP-Induced Gene Expression
Cox-2 inhibition by NS-398 did not affect cAMP-mediated protection. RAW 264.7 macrophages in the presence or absence of 10 µM NS-398 were prestimulated for 15 hr with 1 mM db-cAMP, a combination of LPS (10 µg/ml), IFN-γ (100 U/ml), and NAME (1 mM) (LIN), or remained as controls. Following prestimulation, DNA fragmentation was initiated with 1 mM GSNO and quantitated after 8 hr with the diphenylamine assay described in Materials and Methods. Data are mean values ± SD of three individual experiments (*p ≤ 0.02 versus control samples without NS-398).
