In vitro kinase assay on immunoprecipitated GTK. Cell extracts from RINm5F cells, stably transfected with empty vector (neo-1 and neo-2), Y394-mutated, wild type and Y504F-mutated GTK, were immunoprecipitated with GTK-antiserum and subjected to an in vitro kinase assay for 15 min. The reaction was subjected to SDS-PAGE, and Western Transfer. The phosphorylation was visualised by autoradiography after SDS-PAGE (top) and the total amount of GTK was assessed by Western blotting (bottom). The [32P] incorporation into the myc-tagged GTK was assessed by densitometry and the relative amount was calculated and is indicated below each group.
