Expression of Bcl-2 gene in RINm5F cells (a) and pancreatic islet cells (b), and determination of the transduction efficiency (c). (a and b) Cells were incubated at 37° C for 60 min with different concentrations of adenoviruses. bcl-2 gene expression was induced immediately after infection with different amounts of RU 486. One to 7 or 30 days after transduction, cell lysates were analyzed by Western blot for the expression of Bcl-2 as described in Methods. (c) RINm5F cells (left panel) and islet cells (right panel) were incubated for 60 min in the presence of 30 PFU/cell AdCMVProg and AdG5Trip-Bcl-2 and gene expression was induced immediately after infection with 1nM RU 486. Twenty-four hours (RINm5F cells) or 48 hr (islet cells) after transduction cells were stained for insulin and for Bcl-2 expression as described in Methods and analyzed by flow cytometry. Nontransduced cells (filled histogram), transduced but not induced cells (gray histogram), transduced and induced cells (black histogram).
