Regulation of type I and type III collagen synthesis in BALBc/3T3 fibroblasts by hIL-1a and indomethacin. Cells were preincubated for 48 hr without (0) or with increasing concentrations of IL-1α between 0.01 and 1.0 nM in the absence (w/o) or presence (+) of indomethacin at 10−6, as indicated. Proteins were then biosynthetically labeled with [3H]proline in the presence of ascorbate and β-APN for 24 hr and electrophoresed on SDS-5% polyacrylamide gels (A) with reduction to separate procollagen chains and FN or (B) without reduction to separate pepsin-resistant collagen chains. Migration positions of collagen chains were identified using [14C]-labeled rat tail tendon collagen as a standard on each gel.
