Shb-protein expression in transgenic and control CBA islets. (A) Expression of Shb in islets isolated from Shb-transgenic and control CBA mice. Islets were isolated and dissolved in SDS sample buffer before boiling and electrophoresis and Western blot analysis for Shb protein as previously described (6). Each lane represents the protein extract of about 50 islets from one animal. Equal amounts of protein were electrophoresed in the lanes. (B) CBA islets (1,2) or RIP-Shb islets (35) were pretreated for 30 min at 37°C with 20 µM N-acetyl-leu-leu-norleucinal prior to electrophoresis and Western blot analysis as in A. Each lane shows the extract of 20 islets. Equal amounts of protein were electrophoresed in lanes 1–5. (C) CBA or Shbtransgenic islets from lineages A and B were pretreated, lysed, and subjected to Western blot analysis as in B. Lysate from 20 islets was electrophoresed in each lane.
