Expression of human DAF in transgenic rat tissues. Frozen sections (5 µm) from transgenic (hemizygous DAF+/−rat from line 52.2) or wild-type (WT) rat tissues were fixed in acetone for 10 min at room temperature, air dried, and stained using an indirect immunoperoxidase technique. (A) Immunostaining of serial tissue sections from kidney or heart was performed with an anti-human DAF MAb as first antibody (magnification, ×200). Negative controls were performed using an isotype-matched irrelevant MAb (data not shown). (B) Immunostaining of DAF+/−transgenic rat heart with human anti-Galαl-3Gal antibodies or an anti-human DAF MAb as first antibody followed by staining with a peroxidase-labeled anti-human or anti-mouse Ig, respectively (magnification, ×400).
