FACS analysis of in vitro human complement deposition on wild-type versus transgenic rat ECs. Cardiac endothelial cells were isolated from wild-type (dotted line) and transgenic, either DAF^+/− hemizygous (plain line) or DAF^+/+ homozygous (bold line), rats. Immunostaining was performed on ECs incubated with human serum (dilution 1/10) for 30 min at 37°C. After incubation, ECs were washed and then incubated with either an FITC-labeled anti-human C3, C4, or factor B MAb or with an anti-human C5b-9 polyclonal rabbit serum followed by an FITC-labeled anti-rabbit Ab.
