Electrophoretic mobility shift assay (EMSA) of nuclear extracts confirms loss of NF-κB activity in ischemic cortex, followed by up-regulation at 5 days after onset of ischemia. Cortical nuclear extracts were prepared as described in Figure 3 (see scheme). Moderate levels of constitutive NF-κB activity were observed in sham-operated and contralateral cortical tissues (C). Permanent MCAO induced significant loss of NF-κB DNA-binding at 24 hr. At 5 days after the onset of ischemia, the level of NF-κB activity was significantly increased within the surrounding infarct areas (S) and, to some extent, within the ischemic zone (I).
