Inactivation of SMS1 inhibited BMP2-induced Smad1/5/8 and p38 phosphorylation. The cellular phosphorylation levels of Smad1/5/8, ERK1/2, and p38 were determined by Western blot analysis of femoral osteoblasts from SMS1f/f;SMS2−/− and ERT2-Cre;SMS1f/f;SMS2−/− mice. After treatment with and without 1 μM 4-OHT for 1 day, calvarial osteoblasts were serum starved for 12 h before BMP2 (50 ng/ml) stimulation for the indicated times, and then immunoblotted with antibodies specific to phospho-Smad1/5/8, phospho-ERK1/2, and phospho-p38
