TGF-ß activity assays of cell culture supernatants from Rab31 overexpressing and vector control cells. A Outline of the human TGF-ß activity assay. Concentrated supernatants (1:16) from CAMA-1 Rab31 overexpressing and vector control cells, were added to murine TGFB1−/− fibroblasts transfected with a TGF-ß reporter plasmid. This plasmid encodes secreted alkaline phosphatase (SEAP) under the control of SMAD-binding elements (SBE). SEAP activity is then determined by a chemiluminescent reaction.
