(A) Blood vessels consist of an endothelial cell layer not expressing TF (intima; 1), and TF-containing media and adventitia, indicated by 2. Upon vessel rupture (B), TF comes into contact with the bloodstream and binds the zymogen factor VII. After conversion of FVII to FVIIa, the latter enzyme converts FX to FXa, resulting in subsequent thrombin formation (C) and fibrin deposition.
