Translocation of HMGB1 from nucleus to cytosol is associated with diminished AMPK activation in LPS-stimulated macrophages. (A) Representative images show time-dependent accumulation of HMGB1 in the cytosol after culture of RAW 274.7 macrophages with LPS (100 ng/mL) for 0, 2 or 4 h. (B) Western blots show amounts of HMGB1, phospho-and total AMPK, and tubulin or HDAC in whole cell extracts as well as cytosolic or nuclear fractions obtained from RAW 264.7 macrophages incubated with LPS (100 ng/mL) for 0, 1, 2 or 4 h. (C) Mean band optical density of phospho-AMPK/AMPK and total HMGB1 in cytosolic fractions of macrophages treated as described in (B). Data are means ± SD. Two independent experiments provided similar results.
