HMGB1-induced TNF-a secretion in RAW264.7 cells and blockade of TNF-α secretion by esRAGE and D6-esRAGE. (A) RAW264.7 cells were stimulated with the indicated concentration of HMGB1, and ELISA was used to measure the level of TNF-α secreted into the conditioned medium. TNF-α was secreted in a concentration-dependent manner for 24 h after stimulation with HMGB1, and TNF-α level peaked at 6 h and then tended to decrease. Each point with a bar represents the mean ± SD of three experiments. (B) RAW264.7 cells were treated with the indicated concentration of esRAGE or D6-esRAGE for 1 h, and the cells were then treated with 3 µg/mL HMGB1 for 6 h, followed by measurement of TNF-α level in the conditioned medium.
