Activation of α7 nAChR enhances TGF-β signaling in a PTP1B-dependent manner. (A-C) Effect of knockdown of (A) Ptpn1 on (B) Colla1 and (C) Acta2 mRNA in GTS-21 pretreated rhTGF-β1-stimulated fibroblasts. The fibroblasts were first infected with pLKO.1 scrambled or shRNA-Ptpn1 lentivirus and then pretreated with GTS-21 (6.25, 12.5 and 25 µM separately; PBS was applied in the control group) and stimulated with rhTGF-β1 (5 ng/mL). The cells were harvested 24 h after rhTGF-β stimulation. (D) Effect of knockdown of Ptpn1 on expression of p-Smad2/3, α-SMA and collagen 1 in GTS-21 pretreated rhTGF-β1-stimulated fibroblasts. Using the same experimental setting as Figure 8A, the fibroblasts were harvested 24 h after rhTGF-β1 stimulation and lysed for immunoblotting. Experiments were repeated three times.
