Inflammatory mediators attenuate Sirt6 protein expression in mouse and human colonic epithelial cells. (A, B) YAMC and (C) HT-29 cells were subjected to treatments as indicated. After 24 and 48 h, respectively, total cellular proteins were extracted from harvested cells. Levels of Sirt6 protein were accessed with Western blot using anti-Sirt6 antibody, as described in Materials and Methods. Upper panels illustrate the autoradiographs of representative immunoblots; lower panels show densitometric analysis of the immunoblot data (normalized to β-actin). The experiments were performed three times. Results are expressed as mean ± SEM. n = 3 in each group. *, P < 0.05 versus control; **, P < 0.0l versus control; ****, P < 0.000l versus control.
