Mutant MDM2 proteins are unable to inhibit p53-dependent transcription in fibroblasts

Saos-2 cells were transiently transfected by electroporation with a p53 expression plasmid (pRC/p53), with a p5 3-dependent luciferase reporter plasmid (BP100-GL2), and with either an empty vector or with wild-type or mutant Mdm2 expression plasmids as indicated. The luciferase activity in the various cell lysates was quantitated. Shown is the average of three separate experiments and the standard error of the mean. Only the wild-type MDM2 protein was able in inhibit p53-dependent transcriptional activation.
