[33P]FKBP12 autoradiography of caput and cauda sperm. Caput and cauda sperm were isolated from the epididymis and incubated in rat fertilization medium (RFM) containing [33P]FKBP12 lor 2 hr at 37°C. The sperm were then washed with RFM, wet-mounted onto Superfrost plus slides, air dried, and apposed to NB-2 emulsion for a 2-week exposure. (A) Caput sperm are labeled by [33P]FKBP12. (B) The labeling of caput sperm by [33P]FKBP12 is specifically blocked by the addition of 1 µM FK506 to the incubation medium. (C) Cauda sperm are completely negative.
