Luteolin and quercetin induce cell-cycle arrest followed by death in L. donovani promastigotes. L. donovani promastigotes (5 × 106/ml) were incubated with DMSO (0.2%), 12.5 µM luteolin or 45.5 µM quercetin (dissolved in DMSO) at 22°C, and analyzed for DNA content by flow cytometry as described in “Materials and Methods.” DNA content of T-cell blasts was analyzed in the same way, except that PHA (5.0 µg/ml) was induced in normal human peripheral blood mononuclear cells culture and incubation was carried out at 37°C. Gates were set to assess the percentages of dead (<2n DNA), G0/G1 (2n DNA), and S + G2 + n (>2n DNA) cells. PI, propidium iodide; PBMC, peripheral blood mononuclear cells; DMSO, dimethylsulfoxide.
