Comparison of Amaryl and glibenclamide for efficiacy at the β cell KATP and hypoglycemic activity. The Kd (6.8 nM) and association rate constant, Kon (0.5 min−1/nM−1), values for binding and the inhibitory concentrations (IC50) for depolarization (27.3 nM) determined for membranes of cultured rat insulinoma m5F (RINm5F) cells or intact cells, respectively, were set at 1 for Amaryl, the dose required for equivalent blood glucose decrease in humans was set at 1 for glibenclamide.
