Stability of SMN and SMNc mRNAs in control and interferon (IFN)-treated A172 cells. A172 cells pretreated with medium alone (Control), IFN-β (1000 U/ml), or IFN-γ (1000 U/ml) for 8 hr were subsequently treated with 25 µg/ml 5, 6-dichlorobenzimidazole riboside (DRB) for the indicated time periods, followed by RNA extraction. (A) reverse transcription-polymerase chain reaction (RT-PCR) was performed for SMN, SMNc, and actin genes as described in “Materials and Methods” and the products were analyzed on a 2% agarose gel, or (B) previously digested with Dra I, analyzed on a 7% polyacrylamide gel, and the amounts of SMN and SMNc were quantified and expressed as percentages of initial values measured in the appropriate controls.
