From: SM22α Promoter Targets Gene Expression to Vascular Smooth Muscle Cells In Vitro and In Vivo

Fig. 3

Promoter activity. (A) Comparison of the activity of adenovirus AdSM22α-hpAP and AdCMV-hpAP control virus in primary cell cultures of porcine aortic smooth muscle cells (PASMC), endothelial cells (PEC), and murine fibroblasts (NIH9 3T3). The cells were infected with 250, 500 and 1000 MOI of either AdSM22α-hpAP or AdCMV-hpAP. Percentage of cells expressing AP activity was quantitated 48 hr after infection. Data are expressed as the mean percentage of cells expressing AP activity 6 standard error of the mean (SEM). (B) Comparison of chloramphenicol transferase (CAT) activity in the PASMCs infected with either AdSM22α-CAT or AdCMV-CAT control vector in vitro. CAT conversion activity was quantitated 48 hr after infection. Level of activity between CMV and SM22α promoters was compared in each group by analysis of variance. *p, 0.0001. hpAP, human placental alkaline phosphatase; CMV, cytomegalovirus.
