Quantitative cell-binding assay for scFv MO36 (A) and scFv MO33 (B). Bacterial lysates of scFv MO36 and partially purified scFv MO33 were incubated in different dilutions on 2 × 104 cells (hFAP+ or mFAP+). For subtraction of unspecific binding, all dilutions were also incubated on nontransfected control cells. For detection anti-c-myc mab 9E10, POD-conjugated anti-mouse IgG-Fc detection antibody and TMB substrate (measured at 630 nm) were used.
