Determination of the apparent affinity of MB MO36 for cell expressed human and murine FAP by FACS analyses. Stable hFAP (■) and mFAP (▲) transfectants were incubated with different concentrations of MB MO36 and subsequently with an anti-c-myc mab (9E10) and a FITC-labeled anti-mouse IgG-Fc specific serum. For each dilution, specific fluorescence intensity of 10,000 cells was determined. Mean fluorescence intensity (arbitrary units) is plotted over minibody concentration and apparent affinity is approximated from the determined concentration yielding half maximum binding to each of the transfectants.
