HO-1 activity contributes to cytokine release in RAW 264.7 macrophages. RAW 264.7 macrophages were treated for 8 hr with LPS (10 µg/ml)/IFN-γ (100 U/ml) in the presence or absence of the HO-1 inhibitor SnPPIX (50 µM). After 8 hr, medium was removed and fresh medium containing the same stimulants (LPS, IFN-γ ± SnPPIX) was added to the cells for additional 2 hr. Subsequently, after 2 hr of incubation, conditioned media were removed and analyzed by ELISA for the presence of TNF-α (A) or IL-1β (B). Data are expressed as mean concentration ± SD (n = 3). **p < 0.01 compared with control. #p < 0.05 compared with the conditions as indicated by the brackets.
