A point mutation in the RING finger domain of MDM2 abolishes specific RNA binding

(A) MDM2 expressed from either the PCR clone, which contains the point mutation, or the P2 clone were tested for RNA-binding activity with the Round 10 RNA. Immunoprecipitations were performed from the indicated Sf9 cell lysates with the 419 negative control mAb or the 4B2 and 2A10 MDM2 mAbs. (B) Amino acid sequence comparison of the C-terminal 68 amino acids of HDM2 and MDM2. Identical amino acids are indicated by an asterix and conservative amino acid differences by a period. Numbers indicate amino acid positions of HDM2 and MDM2. A G446S mutation in MDM2 abolishes its ability to bind specifically to Round 10 RNA.
