Construction of pTfRscFv, the expression vector for the lipid-tagged anti-human transferrin receptor scFv (TfRscFv). The vector pLP1 contains an amino acid linker sequence between the E. coli lipoprotein signal peptide and the scFv cloning site (LPP and linker), and also contains both c-Myc and His6 tag sequences (myc and His). The PCR-amplified TfRscFv cDNA from the vector pDFH2T-vecOK was cloned into pLP1 using NcoI and NotI sites to obtain the expression vector, pTfRscFv.
