Effect of AP-1 and the SPR1 inhibitory element on transcriptional activity. CAT constructs with wild-type AP-1 and altered sequences flanking the AP-1. CAT assays were performed in 70N normal breast cells transfected with pBLCAT2 vector (lanes 1,2); an oligonucleotide containing a wild-type AP-1 site with flanking sequences completely different from those in the SPR1 promoter (pBLCAT2/APl WT2-3; lanes 3,4); the SPR1 wild-type AP-1 site and the inhibitory element (pBLCAT2/INH AP1; lanes 5,6); and the SPR1 wild-type AP-1 site (pBLCAT2/AP1 WT1-1; lanes 7,8). Extracts of 15 β-Gal units were assayed for CAT activity. Results are typical of duplicate experiments.
