Effects of insulin on PTEN phosphorylation in GFP-RIN and GFP + Shb-RIN cells. GFP-RIN or GFP + Shb-RIN cells (2 × 105) were labeled for 60 min with 32P-phosphate and then stimulated for 10 min with insulin (10 µg/ml). PTEN was immunoprecipitated, electrophoresed on a 9% SDS-PAGE, and transferred to an Immobilon filter, which was exposed to x-ray film. Nonprecipitated lysates were run in parallel. The autoradiographic bands shown in (A) were quantified and expressed per total protein 32P-incorporation (B). Results are means ± SEM for three to four observations. Comparisons were made using one-way ANOVA for repeated measurements (p < 0.05). Individual differences were obtained using Student Newman Keuls test. *p < 0.05 versus corresponding control cells.
