SNOC-mediated inhibition of IL-2 mRNA expression is reversible. EL4-6.1 cells were incubated without (lane 1) or with 1000 U/ml IL-1β in the absence (lane 2) or presence of 1 mM of SNOC (lane 3) or SNOC−NO (lane 4). After 3 hr, cells were carefully washed and incubated for 24 hr in fresh culture medium. Cells were then incubated again without (lane 1) or with IL-1β (lanes 2–4) for 3 hr. IL-2 mRNA was quantitated in relation to GAPDH mRNA. Under these conditions, inhibition of IL-2 mRNA expression was fully reversible. Values are mean ± SD of three independent experiments.
