Increased transgene expression in μΜΤ mice and repeated dosing of adenoviral vectors in μΜΤ and C57BL/6J mice. (A) hAAT levels in μMT and control C57BL/6J mice at 5 and 19 days after initial intravenous administration of AdhAATΔE1 at a dose of 2 × 109 PFU/mouse (4.4 × 1010 particles); n = 5 for each group. (B) Histological sections from livers of C57BL/6J and μMT mice treated at day 1 with either no vector or with AdhAATΔE1, as described in Fig. 1A and then treated on day 21 with AdβgalΔE1 at 3 × 109 PFU/mouse (3 × 1010 particles). Blue staining indicates color development dependent upon the presence of the β-galactosidase transgene. Animals were sacrificed 3 days after AdβgalΔE1 injection. Representative samples from 3 mice in each group are shown at low magnification.
