IFN- α treatment extinguishes the DNA-binding activity of constitutively activated Stat1 and Stat3 in melanoma precursor lesions.

(A) Protein extracts (20 µg/sample) of sections of atypical nevi, obtained from a patient before (a and b) and after treatment (c and d), were incubated with radiolabeled hSIE probe and subjected to EMSA. (B) Protein extracts (20 µg/sample), prepared from sections of an atypical nevus (a–c) and normal skin surrounding the nevus (d–f), obtained from a patient before IFN-α treatment, were incubated with a Stat1 (a and d), Stat2 (b and e), and Stat3 (c and f) antibody, followed by incubation with the radiolabeled hSIE duplex oligonucleotide and EMSA.
