Time-dependent appearance of fibrocytes in cultures of adherent, human peripheral blood leukocytes

Peripheral blood leukocyte fractions were isolated as described in Materials and Methods, cultured on glass cover slips, fixed, and double-stained with rhodamine-conjugated monoclonal anti-human CD34 antibody plus FITC-conjugated monoclonal anti-human collagen I antibodies. Similar results were obtained by double-staining with anti-CD34 plus monoclonal anti-vimentin antibodies. At least 10 positive cells were enumerated within each field (mean ± SEM of ≥3 fields/time point).
