Tan I had no impact on NLRC4 or AIM2 inflammasome activation. (A–C) LPS-primed BMDMs were incubated with or without Tan I (20 µM), followed by the indicated stimuli. Cleaved caspase-1(p20) and mature IL-1β (p17) in supernatant were assessed by immunoblotting (A), activity of caspase-1 (B) and IL-1β secretion (C) in the supernatants were analyzed. (D-F) LPS-primed BMDMs were incubated with or without Tan I (20 µM), followed by treatment with nigericin, poly(dA:dT) or Salmonella, or Pam3CSK4-primed BMDMs were treated with Tan I followed by transfection of LPS, cleaved caspase-1(p20) and mature IL-1β (p17) were assessed by immunoblotting (D), activity of caspase-1 (E) and IL-1β secretion (F) in supernatants were evaluated.
