Tan I inhibited NLRP3 inflammasome by disrupting the association between NLRP3 and ASC. (A) BMDMs were primed with LPS followed by Tan I incubation, and then treated with nigericin, the efflux of potassium was measured. (B) The mobilization of Ca2+ in LPS-primed BMDMs treated with Tan I (or not) followed by ATP for the indicated time was assessed. (C) LPS-primed BMDMs were cultured in the presence or absence of Tan I and then stimulated with nigericin, then stained with MitoSox, mtROS was analyzed by flow cytometry. (D, E) Immunoprecipitation analysis of the interaction among NLRP3, NEK7, ASC in HEK-293T, in the presence of Tan I (20 µM) or not.
