The cytotoxic effect of A30P-aSyn-O to primary dopaminergic neurons in culture is mediated by calcineurin. Mouse mesencephalic neurons were treated with 5 µM of different A30P-aSyn species (monomer, oligomer of fiber) in the absence or in the presence of 2 µM of CsA. After 24 h of treatment, cell viability was evaluated by LDH release (A). The number of neurons per field was quantified (B) in the cultures treated with A30P-aSyn-O in the presence or absence of CsA, and immuno-stained with anti-MAP2 antibody (C).
